The realisation of the main project demands a reliable tool for the specific visualisation of the investigated GAl4 expression patterns as well as a global visualisation of synapic neuropil as a reference for the localisation of the expression patterns. A well established tool for the unspecific staining of synaptic neuropils in Drosophila melanogaster is a immunohistochemical approach with the antibody nc82 (A.Hofbauer,1991). This has been adopted from earlier works in our lab (K.Rein,2002 ). For the best visualisation of the Gal4 expression patterns we are looking for a reporter line with a reliable overall GFP expression in the cellpopulation defined by the Gal4 line. In this approach we crossed several Gal4-lines with eleven different reporter lines and scanned the brains of the offspring with a laser scanning confocal microscope. The aim of this is to reveal the correlation between the reproter lines construct and the resulting expression patterns. so we choose to compare the different expression pattern in multiple aspects:

  • Specifity and power of the construct
  • Dependency of the expression patterns to the construct
  • Dependency of the expression patterns to the localisation of the construct on different chromosomes
  • Genderspecifity of the expression patterns
